Tetracycline detection in milk samples using a non-radiative fluorescence sensor of molecularly imprinted polymer nano-sheets#

摘要:四环素是广泛用于人、畜禽与水产养殖的广谱抗生素,过量长期使用会加剧微生物耐药性。本研究采用一锅溶胶-凝胶聚合制备荧光分子印迹聚合物纳米片(NS-FMIPs),将其作为光学传感器实现四环素快速选择性检测。传感器306 nm激发下于410 nm产生荧光,传感机理为四环素与NS-FMIPs之间的荧光共振能量转移(FRET)诱导淬灭。通过XRD、FTIR、FESEM、HRTEM、XPS、紫外-可见光谱、TGA完成全面材料表征。探针对四环素选择性优异,1 min内即可观察到荧光淬灭,其他受试药物几乎无响应。优化条件下,5–60 nM浓度区间线性良好,R²=0.9948,检出限2.64 nM,印迹因子4.137,灵敏度高。在牛奶与水样中完成实际样品验证,回收率78.4%-97.5%。该NS-FMIPs传感器制备简便、选择性与灵敏度高,可在复杂基质中实现四环素精准监测。

Dual-functional fluorescence probe with well-separated channels for evaluating bisulfite/viscosity in biosystem and food samples#

摘要:亚硫酸氢盐因具备抗菌、漂白、抗氧化性能,广泛应用于饮用水与食品加工,但过量摄入与多种不良健康结局相关,亟需稳定灵敏的检测手段。本文设计双功能荧光探针CSO-SN,以硫代巴比妥酸为受体、噻吩香豆素为给体,实现亚硫酸氢盐与粘度双通道互不干扰检测。探针对亚硫酸氢盐响应时间低至10 s,检出限0.235 μM,抗干扰能力优异;高粘度环境下产生近红外荧光信号,荧光对数与粘度对数具备良好线性关系。在活细胞与斑马鱼模型中均验证传感响应,在生物体系、食品安全、环境监测领域具备实际应用潜力。

A long-wavelength and color-changed cellulose-based fluorescent sensor for Al3+ and its applications in detection and fluorescence imaging#

摘要:通过3-羟基-4-(2-羟基-4-磺-1-萘偶氮)萘-2-羧酸与乙基纤维素反应,合成纤维素酯荧光衍生物EC-HNCA。利用¹H NMR、FT-IR、XRD系统表征合成前后结构变化。EC-HNCA溶液在365 nm紫外灯下本底无荧光;加入Al³⁺后产生明亮粉红色荧光,对Al³⁺选择性突出。610 nm峰值荧光强度与0–6.5 × 10⁻⁶ mol/L的Al³⁺浓度线性相关,R²=0.9910,检出限7.6 nM。EC-HNCA-Al³⁺复合物在不同溶剂中表现多变发光颜色;可用于西瓜果皮维管组织荧光成像,也可对白萝卜、大蒜、毛竹笋、大白菜中的Al³⁺进行定量测定,在植物组织荧光成像与铝离子检测方面具备应用前景。

A quenching-recovery fluorescence assay for discriminating triphenylmethane dyes using sulfur nanodots#

摘要:三苯甲烷(TPM)染料工业应用广泛,种类多、光谱重叠,实现区分识别存在挑战。本文提出基于硫纳米点(S-dots)的荧光传感策略,硫纳米点同时充当荧光指示剂与染料降解试剂,实现碱性品红(BF)、结晶紫(CV)、乙基紫(EV)三种TPM染料鉴别。加入染料后,静态淬灭机制使硫纳米点本征蓝色荧光发生淬灭;在硫纳米点表面S²⁻基团介导下染料逐步降解,荧光发生恢复。通过监测荧光淬灭-恢复动力学过程,可有效区分三种染料,检出限分别为7.64 μM、11.16 μM、4.79 μM。该方法对常见干扰物耐受度高,加标湖水样品回收率99.1-105.9%;进一步开发便携检测试剂盒,实现现场肉眼快速鉴别。

Advances and future prospects in colorimetric and fluorescence sensors for Pb2+, Hg2+, and Cd2+ ion detection: A review#

摘要:该综述批判性总结Pb²⁺、Hg²⁺、Cd²⁺离子的比色与荧光传感器最新进展,聚焦分子设计与纳米工程策略;对比聚集诱导发光、光诱导电子转移、化学剂量计等传感机理,关联构效关系与性能指标。区别于以往综述,重点讨论实验室探针向现场实用器件转化的现存难点:复杂基质中超痕量检测、共存离子干扰、长期稳定性与可重复使用性。指出真实水体选择性不足、货架寿命有限、标准化测试方案缺失等关键短板,提出面向现场重金属检测传感器开发的战略性发展方向,弥合基础科研与实际环境监测之间的鸿沟。

A three-channel fluorescence sensing array based on a multi-layer perceptron for precise discrimination of eight common metal ions#

摘要:多种金属离子共存威胁生态环境与公共安全,亟需快速、高通量、高分辨检测手段。相似理化性质离子在复杂基质中难以特异性识别,本文构建三通道荧光传感阵列结合深度学习智能识别策略。制备Lap-Eu-PEF、Lap-Eu-MOX、Lap-Tb-PEF传感单元,采集八种金属离子不同浓度下的颜色信号,构建高维特征数据集。利用线性判别分析(LDA)降维可视化,提升离子之间可分性;搭建多任务多层感知机(MLP),同步完成离子类别判别与浓度预测。独立测试集上分类准确率100%,浓度预测R²>0.999,平均绝对误差0.18 ± 0.03 μM,模型鲁棒性与泛化能力优异。进一步结合YOLOv5与MLP开发端到端荧光图像智能分析系统,实现图像采集、区域识别、信号提取、定性定量全流程自动化。

From fluorescence sensing to anode protection: A coordinating polyacylhydrazone for zinc ion recognition and long-cycling supercapacitors#

摘要:在同一材料体系同时实现高灵敏离子识别与稳定界面调控,是配位化学与功能材料交叉领域的一大挑战。本文报道一类局域共轭聚酰腙PID-F,依靠配位诱导簇发光(CTE)协同机制实现Zn²⁺特异性荧光识别,Zn²⁺检出限低至0.29 μM,在多种干扰离子存在下表现优异的螯合增强荧光(CHEF)选择性。更重要的是,将PID-F进一步应用于锌离子混合超级电容器(ZHSC)锌负极保护层;PID-F富含N/O配位位点,能够调控Zn²⁺均匀沉积,显著抑制枝晶生长与副反应。1 A g⁻¹电流密度循环11 400圈,Zn@PID-F电极库伦效率高达99.9%,容量保持率84.2%,实现Zn²⁺荧光识别与电极界面防护功能一体化。

A schiff base-functionalized europium metal–organic framework for turn-off fluorescence sensing of Cu2+#

摘要:开发低成本、高灵敏、精准的荧光检测手段对降低工业废水重金属污染意义重大。本文合成席夫碱修饰铕基金属有机框架Eu-MOF,用于Cu²⁺“关型”荧光传感。由2-氨基对苯二甲酸与2-羟基烟醛得到席夫碱配体,再与Eu³⁺配位构筑框架,样品前处理流程简便快速。材料荧光行为具备pH依赖性;2.5-500 mg L⁻¹浓度区间对Cu²⁺线性响应,定量限LOQ=2.5 mg L⁻¹,检出限LOD=0.83 mg L⁻¹。在食醋、茶水、果汁、水样中开展加标回收,回收率84.5%-100.9%,相对标准偏差RSD=1.7%-4.8%,适用于多种基质中Cu²⁺测定。

Selective fluorescence turn-on detection of methylglyoxal using N-substituted NBD derivatives#

摘要:甲基乙二醛(MGO)是参与糖基化、氧化应激、代谢疾病的活性α-二羰基化合物,生物样本中检测难度高。本文设计一系列N-取代-4-肼基-7-硝基苯并呋喃(NBD)衍生物用于MGO荧光检测。在肼基氮原子上引入烷基,调控探针反应活性、位阻可及性以及生成腙的荧光性质。对比未取代NBD-H,多数N-取代衍生物对MGO响应更强、产物更稳定,对竞争羰基化合物响应更弱。色谱与质谱证实探针-MGO腙类产物生成;理论计算支持探针优先在MGO的醛基位置发生缩合。在人血浆体系中,即便存在乙二醛、丙烯醛、乙醛、甲醛等干扰羰基,多数衍生物仍保持强线性荧光响应,分析灵敏度显著提升,仅需蛋白沉淀步骤即可适配微孔板高通量检测。

Fluorescence aqueous detection of CO2 via protonation of 2-phenyl-4-methylimidazole#

摘要:二氧化碳浓度对生命生产、农业生长、海洋环境至关重要。相比现有检测方法,亟需开发适用于气相与水相的简便高灵敏CO₂检测方法。本文筛选得到2-苯基-4-甲基咪唑(PMZ)@聚甲基丙烯酸甲酯(PMMA)传感薄膜,适用于水相检测,对CO₂表现高灵敏荧光响应。水相中CO₂溶解形成碳酸,进而使薄膜中荧光物质质子化生成PMZH⁺,诱导荧光发射蓝移。该传感膜响应时间约5 min,检测线性范围1.8%–6.0%,适用于酿造过程控制、农业生产仓储、生命科学孵育、早期安全报警等场景。薄膜制备简便、易于存储,为CO₂传感提供理论基础与实用平台。

Peroxynitrite-activated near-infrared fluorescence sensing platform for early non-invasive IBD diagnosis, herbal formula efficacy evaluation and anti-enteritis bioactive components screening#

摘要:炎症性肠病(IBD)严重影响生活质量、提升结直肠癌风险,早期诊断与及时干预对控制病情至关重要。过氧亚硝基阴离子(ONOO⁻)是IBD发病关键活性氮物种,是极具吸引力的生物检测标志物。本文理性构建靶向ONOO⁻的开启型近红外荧光探针HCY-PO。ONOO⁻触发二苯基磷酰胺识别基团断裂,探针750 nm处荧光显著增强,60 s内响应,检出限90 nM,复杂生物体系中抗干扰能力强。利用该探针实现炎症细胞、斑马鱼肠炎模型内源性ONOO⁻实时追踪,以及小鼠溃疡性结肠炎早期诊断;荧光强度与IBD严重程度正相关。可实现小鼠粪便ONOO⁻灵敏检测,用于IBD进展无创监测。基于该性能,首次建立集成荧光传感平台,半定量评估中药复方补中益气汤疗效,荧光、病理、炎症因子结果一致性良好,并高效筛选出5种抗肠炎活性成分。

A tri-emissive dye-encapsulated white-light fluorescence sensor based on MIL-53(al) for the visual detection of pesticides and metal ions#

摘要:通过一步溶剂热法合成MIL-53(Al),包埋9,10-双(苯乙炔基)蒽(BPEA)与偶氮苯(FG)两种染料,构建三发射荧光传感器BPEA/FG@MIL-53(Al)。调控两种染料比例,实现冷白光发射,CIE坐标(0.31, 0.31),可用于氟菌唑、四氟苯脲、Al³⁺、Fe³⁺检测。与靶标分析物作用后,BPEA/FG与MIL-53(Al)骨架之间发生能量/电子转移,荧光增强或淬灭;氟菌唑检出限0.35 μM、四氟苯脲0.21 μM、Al³⁺ 0.36 μM、Fe³⁺ 0.33 μM。开发配套颜色追踪与传感应用程序,连接手机APP快速采集RGB值,通过曲线拟合计算靶标浓度,实现多目标多模式现场便携检测。

Facile construction of an organic-inorganic PO@Ag₂S/CuS hybrid nanoprobe with GLS-responsive NIR-II fluorescence#

摘要:开发刺激响应、可开关近红外二区(NIR-II)发射的杂化纳米材料,对先进光电子与生物光子应用意义重大。本文通过乳化策略简便制备有机-无机杂化纳米探针PO@Ag₂S/CuS,由Ag₂S与CuS量子点共包载于生物响应型两亲聚(γ-谷氨酸)-十八胺(PGA-ODA)聚合物壳层。完整组装中CuS量子点通过浓度依赖机制作为高效淬灭剂,体系处于荧光“关闭”状态。暴露于谷氨酰胺酶(GLS)后,PGA-ODA壳层被特异性酶降解,诱导纳米探针结构解组装,Ag₂S与CuS空间分离,触发NIR-II荧光显著恢复。系统结构形貌均一、元素分布均匀、光学性能动态可调;荧光成像证实其对GLS的时间、浓度依赖性响应与优异生物安全性。

Dispersive liquid-liquid microextraction coupled with ultra performance liquid chromatography-fluorescence detection for the determination of polycyclic aromatic hydrocarbons in tea#

摘要:建立改进分散液-液微萃取-超高效液相色谱-荧光检测(DLLME-UPLC-FLD)方法,测定马来西亚市售茶叶中8种欧盟食品安全局规定多环芳烃(PAHs)。DLLME以二氯甲烷作萃取溶剂、甲醇作分散溶剂;C18色谱柱分离,270 nm激发、四通道发射波长荧光检测。方法线性良好,检出限3.87-32.5 ng L⁻¹,RSD≤9.91%,回收率85.13-101.71%,富集因子35-48,AGREE绿色度评分0.53。分析43份商用袋泡茶与浸出茶样品,䓛为最主要PAH;∑PAH₈总浓度排序:后发酵茶 > 未发酵茶 > 全发酵茶 > 半发酵茶 > 花草茶;所有品类暴露边际值均高于安全临界阈值10 000。

Mechanistically derived fluorescence rancidity index for real-time monitoring of olive oil oxidation under thermal, photo, and frying conditions#

摘要:开发基于荧光的快速、非破坏性橄榄油氧化监测方法。提出荧光酸败指数(FRI),以新鲜油荧光为基准的比率型光学指标;使用自研532 nm激发激光诱导荧光系统评估特级初榨橄榄油、果渣橄榄油,分别开展热氧化180 ℃、紫外氧化、反复煎炸处理。氧化过程中叶绿素相关发射(~675 nm)逐步衰减同时发生蓝移。热氧化条件下FRI与PAV(r=0.92)、TOTOX(r=0.91)、INTOX(r=0.92)、CDV(r=0.88)强相关;紫外氧化条件无显著相关性;反复煎炸过程FRI持续上升。FRI是快速无试剂橄榄油氧化光学指标,尤其适合热氧化场景,但响应行为依赖氧化途径。

A fluorescence aptasensor for high sensitivity detection of golgi protein 73 based on N, S-GQDs and MoS₂@RGO via fluorescence resonance energy transfer mechanism#

摘要:高尔基体蛋白73(GP73)是极具前景的肝细胞癌(HCC)生物标志物,准确定量对早期诊断至关重要。本文构建荧光适体传感器,基于FRET机理,以GP73适体功能化氮硫共掺杂石墨烯量子点(N, S-GQDs-GP73Apt)为荧光给体,二硫化钼@还原氧化石墨烯(MoS₂@RGO)为荧光受体。不存在GP73时,给体荧光被有效淬灭;加入GP73后,特异性结合触发给体-受体复合物解离,破坏FRET,荧光恢复。优化条件下,0.001-100.0 μg/mL浓度范围线性良好,检出限0.019 ng/mL。人血清样品验证相对误差0.76%-3.85%,与ELISA方法无统计学差异,是可靠低成本的早期肝癌诊断平台。

Triplet State Engineering in Regioisomeric Cocrystals Enabling Dual Phosphorescence and Efficient Thermally Activated Delayed Fluorescence#

摘要:有机给受体共晶是通过超分子组装调控激发态过程的多功能材料。本文报道一系列位置异构D-A共晶(oDCB、mDCB、pDCB),同时表现热活化延迟荧光(TADF)以及来自³CT与³LE态的双重磷光。受体结构变化可系统调控分子间电荷转移相互作用,改变单重-三重态能隙ΔE_ST,进而调控反向系间窜越(RISC)速率。oDCB共晶ΔE_ST最小,RISC速率最快,可达1.29 × 10⁵ s⁻¹;pDCB拥有最大ΔE_ST(0.23 eV),激发态寿命最长为2.12 ms,RISC速率最慢(6.97 × 10² s⁻¹)。77 K条件下,全部共晶在振动耦合协助下产生来自³CT、³LE的双重磷光。共晶三重态能量图景由母体组分三重态能级决定,进一步影响³LE与³CT之间振动耦合程度。

Photoactivation-Enabled Tunable Room-Temperature Phosphorescence and Thermally Activated Delayed Fluorescence in Crystalline Materials#

摘要:晶态材料分子堆积刚性强,很难实现光激活可调控室温磷光(RTP)与热活化延迟荧光(TADF)。本文首次在无需预处理的晶态体系中实现本征光激活RTP,并且通过合理主客体工程实现TADF的开启-关闭切换。将二萘并呋喃(OA)嵌入三种晶体宿主,构建普适晶态光激活平台。实验与计算表明能级匹配与激子寿命主导RTP/TADF调制:二苯并呋喃(DBF)宿主依靠主客体能量转移,同时产生RTP与TADF;4-(甲氨基)苯甲酸甲酯(MAB)、硼酸(BA)宿主因为能级失配或者激子快速淬灭抑制TADF,但依靠高结晶度与刚性氢键网络促进RTP发射。机理分析证明紫外光照调控激发态载流子浓度与跃迁概率,从而实现光激活,在时间分辨防伪领域具备应用潜力。

ct DNA-Driven Helical Assembly of Achiral Dimers: Simultaneous Activation of Three-Photon Fluorescence and Circularly Polarized Luminescence#

摘要:同时具备圆偏振发光(CPL)与高阶多光子激发荧光的有机材料,对近红外成像、生物手性微环境传感意义重大。本文提出简便手性组装策略,借助小牛胸腺ct-DNA与非手性吡啶鎓同源二聚体相互作用,同步实现CPL与三光子激发荧光(3PEF)。精确设计两种空间构型不同的同源二聚体LD、FD;线型LD分子厚度窄,依靠静电相互作用优先结合ct-DNA小沟,实现从DNA骨架向小分子高效手性传递,同时有效抑制非辐射衰减,触发三光子荧光,最终输出显著CPL信号。本工作提供可与核酸适配的非手性小分子设计思路。

Aggregation-Enhanced Photocleavage for Time-Resolved Multicolor Fluorescence Encryption and Quaternary Optical Storage#

摘要:时间分辨多色动态荧光系统在高级信息加密与光存储领域需求旺盛,但多数体系光化学动力学固定或组装复杂。本文报道聚集调控光化学体系,通过聚集依赖激发态工程实现光裂解动力学连续编程。理性设计给受体发光体TPEMN,中心乙烯桥可发生光裂解。光照射下,聚集增强光氧化裂解过程,反应速率随聚集程度逐步加快,伴随荧光从橙色到蓝色连续演化。实验与理论表明,聚集抑制激发态结构弛豫、延长激发态寿命、促进系间窜越、加速活性氧生成,共同加快光裂解动力学。将传统二元光响应转化为时间分辨多色荧光态,可实现带干扰项的时间门控信息加密、“读后即焚”多级防伪、高密度四元光学存储。

Luminescence thermometry of LiYF₄: Tm³⁺ down-conversion phosphor using the fluorescence intensity and emission color#

摘要:首次研究LiYF₄⁺的温度依赖下转换发光。在203 K至603 K宽温区开展系统磷光基传感研究。采用微波燃烧法快速、高效、低成本合成LiYF₄% Tm³⁺微晶粉末;分析结构、形貌与发光性能,建立掺杂浓度与光学响应关系。0.6 mol% Tm³⁺掺杂样品发射强度最高,且发射强度随温度升高表现反常热增强,源于热辅助的Tm³⁺能级布居重分布。首次在该体系实现双模式发光测温,基于荧光强度与CIE色度坐标实现精准温度传感;203 K FI模式下最大相对灵敏度达1.14% K⁻¹,是首个基于下转换发射的LiYF₄⁺光学温度计。

Spatiotemporal Sequential Fluorescence and Electrochemistry Bimodal Quantitation of Enzyme Activity in a Living Cell#

摘要:活细胞内酶活性的时空精准单细胞定量,对理解细胞异质性、疾病筛查至关重要。本文开发时空序列荧光-电化学双模态平台,实现活单细胞中天冬酰胺内肽酶(AEP)的空间成像与精确定量。通过θ纳米吸管将双AIE探针pyTPE-CBT递送进单细胞,荧光增强11.18倍,实现AEP空间分布可视化。荧光成像引导下,将纳米吸管检测通道定位至目标亚细胞区域;AEP切割探针分子,改变电荷密度与有效通道尺寸,调制离子电流实现定量检测,检出限LOD=3.83 ng/mL。该方法克服传统单模式检测缺陷。

AI-driven dual-mode phase and label-free fluorescence imaging platform using a single-shot gabor hologram#

摘要:同时获取定量相位与荧光信息对全面细胞分析至关重要,但传统双模态系统需要标记、复杂光学与多路采集。本文搭建AI驱动双模态相位-无标记荧光成像平台,仅依靠单次Gabor全息图,同时重建定量相位图像与虚拟荧光通道,无需荧光染色、多次拍摄、多模态硬件。引入单向无监督扩散模型,学习从全息图向双模态输出的单向映射;结合对比学习全息合成与全息条件去噪扩散,保证高结构保真度与稳健跨模态重建。在多种癌细胞系、细胞器特异性通道验证,可准确还原细胞形态与亚细胞分布;FID=57.74,SSIM=0.76,PSNR=26.89 dB,LPIPS=0.12,对高放大倍率、不同细胞类型、低光照、离焦样本具备泛化能力。

Using fluorescence lifetime imaging microscopy to probe the metabolic peritumor microenvironment and beyond#

摘要:荧光寿命成像显微镜(FLIM)可实现无标记非破坏性光学代谢成像,已成为多尺度解析肿瘤微环境(TME)的主流平台。但肿瘤周边的瘤旁微环境(pTME)研究相对匮乏。本篇综述介绍FLIM用于肿瘤代谢微环境研究现状,以及在瘤旁微环境中的应用进展。在TME领域,FLIM充分利用代谢异质性表征各类肿瘤细胞状态,用于体外/体内药物筛选,区分肿瘤与瘤旁组织。相比之下,仅有少量体外工作探究pTME,提示肿瘤-瘤旁交互会改变瘤旁细胞代谢。多数诊断研究将瘤旁直接归类为正常组织,忽视其促肿瘤作用。评估pTME对肿瘤进展与治疗响应的贡献,是FLIM领域值得挖掘的方向。

Fluorescence-guided surgery for necrotizing soft tissue infections: A systematic review#

摘要:坏死性软组织感染危及生命,需快速外科清创,依靠肉眼识别活组织。荧光引导手术越来越多地用于评估组织灌注,有望改善诊疗。遵循PRISMA与Cochrane规范开展系统综述,共检索到237条记录,最终纳入2项病例研究共3例患者。两项研究均证明该技术在清创术中具备可行性,无不良事件报告。现有证据等级不足,无法得出确定性结论;但提示ICG荧光引导手术具备评估组织灌注的潜在价值,亟需更多研究。

Intraoperative indocyanine green fluorescence angiography for flap perfusion assessment in head and neck oncological reconstruction: A systematic review#

摘要:评估术中吲哚菁绿荧光血管造影(ICG-FA)在头颈部肿瘤重建皮瓣灌注评估中的临床价值。纳入10项观察性研究共463例患者、469块ICG评估皮瓣;2项对比研究306块对照皮瓣。绝大多数为游离皮瓣(90.6%)。ICG评估皮瓣重返手术室占12.4%,部分坏死4.8%,全部丢失3.8%;受损皮瓣挽救率73.3%;12.8%病例因成像改变术中处理。ICG-FA可作为术中灌注评估的有效辅助手段,在皮瓣挽救、降低皮瓣丢失、辅助手术决策方面具备潜在收益;但证据受限于观察性设计与异质性,需要前瞻性对照研究才适合常规推广。

A fluorescence-based high-throughput assay enabling engineering of BsPPS, a unique phenolic compound–phosphorylating enzyme from Bacillus subtilis#

摘要:能够磷酸化酚类化合物的酶体系稀缺;枯草芽孢杆菌酚磷酸合成酶(BsPPS)可区域选择性磷酸化多种天然多酚,对提升植物化学物水溶性与生物利用度极具价值。但缺少高效高通量筛选平台限制其改造与应用。本文以伞形酮为荧光探针,开发基于荧光的高通量检测方案,快速评估BsPPS酶活。伞形酮发生磷酸化会引发显著荧光淬灭,荧光强度与催化活性呈负相关。整套检测体系经过系统优化验证,准确度、精密度、重现性良好。应用于易错PCR突变文库,筛选得到热稳定性提升同时保留催化活性的BsPPS突变体。

DNA-Based Loop-Amplifier Fluorescence Signaling Networks Enable Rapid Quantitative Pathogen Detection of Periprosthetic Microbial Bone Infection#

摘要:假体周围微生物骨感染(PMBI)病情凶险,延误治疗后果严重,临床难点在于病原微生物高效精准识别。本文构建基于DNA的环路放大器荧光信号网络(DLFSN)用于PMBI病原体检测。DLFSN包含放大器单元A与定量单元B:放大器单元A依靠熵驱动自动反应环路,将病原体遗传物质转化为荧光DNA分子,高效放大靶标基因组信号;定量单元B将淬灭态荧光中间体转为激活态,实现病原遗传特征精准定量。DLFSN诊断准确度与组织切片染色、qPCR等金标准相当,真实临床样本总检测时长仅约30 min,为PMBI早期诊断提供有前景的精准检测工具。

A pH-Responsive fluorescence lifetime probe reveals GOT1 phase separation in cellular anti-senescence#

摘要:细胞衰老伴随进行性酸化,但细胞如何感知并适应pH变化仍不清楚。本研究发现代谢酶GOT1作为pH传感器,发生液-液相分离(LLPS)以对抗衰老。蛋白质组学发现衰老的人肺细胞中GOT1上调;模拟衰老的酸性条件直接通过N端内在无序区(IDR1)诱导GOT1发生液液相分离,招募ME1形成动态酶促凝聚物,清除活性氧、减轻氧化应激。设计BDP-PLP荧光探针,结合GOT1辅因子吡哆醛磷酸与BODIPY荧光团,通过结合抑制PET机理,实现高特异性GOT1靶向与pH依赖荧光寿命成像。利用FLIM实现活细胞内GOT1凝聚体pH动态定量实时可视化,揭示相分离产生的强酸性局部微环境是其抗衰老功能的关键。

Intraoperative indocyanine green fluorescence guidance to reduce ureteroenteric anastomotic strictures during robot-assisted radical cystectomy. A systematic review and meta-analysis#

摘要:输尿管肠吻合口狭窄(UAS)是机器人辅助根治性膀胱切除术(RARC)的临床相关并发症,主要由输尿管缺血导致。吲哚菁绿(ICG)荧光被提议用于吻合前识别缺血段。本系统综述与meta分析评估ICG对UAS发生率的影响。纳入4项观察性研究共886例患者、1739根输尿管:369根(41.6%)接受ICG引导评估,517根(58.4%)为标准评估。合并分析显示UAS发生率无统计学显著降低(RR 0.31,95%CI 0.08-1.16;P = 0.08;I² = 76.4%)。排除最高偏倚研究后异质性消除,结果倾向ICG获益(RR 0.20,95%CI 0.09-0.46;I² = 0%),提示合并估计不稳定,受研究异质性影响大。输尿管切除长度、淋巴结阳性、手术时间无显著差异。现有证据局限于非随机研究,偏倚风险高、证据确定性很低,需要前瞻性随机研究明确临床获益。

An Ultra-Sensitive Liquid Chromatography-Fluorescence Detection Method for Determining hCE1 Activity and Evaluation of hCE1 as a Biomarker of Liver Injury#

摘要:人羧酸酯酶1(hCE1)主要在肝脏高表达,肝细胞损伤后释放入血,有望成为肝损伤生物标志物。现有方法选择性差、灵敏度有限,易受多种蛋白干扰。本文设计合成新型荧光探针NCMNe,建立超灵敏液相色谱-荧光检测(LC-FD)方法,定量血浆hCE1活性。特异性实验证明NCMNe可被hCE1选择性水解,不受血清白蛋白、对氧磷酶1、丁酰胆碱酯酶干扰。定量下限0.0001 μM,是目前最灵敏的hCE1活性测定手段。临床样本分析显示乙肝患者血浆hCE1活性显著高于健康对照,与谷草转氨酶、谷丙转氨酶高度相关;ROC分析AUC=0.901,诊断性能与传统肝酶相当。

“Synergistic Effect of CQDs on the Spectroscopic Behaviour and Bioactivity of Azo Dyes: Bio-imaging Applications”#

摘要:偶氮染料类药样性质优异,本身固有荧光使其有望用于生物成像,但强度与量子产率偏低。碳量子点(CQDs)可显著增强染料荧光。本研究假设新合成偶氮染料与CQDs相互作用,通过特定光物理途径实现荧光增强。利用多种光谱手段探究相互作用;测定量子产率、Zeta电位,开展抗菌、抗氧化、溶血等体外评价。与CQDs复合之后,染料荧光强度提升、激发态寿命延长、量子产率改善;增强机理来自FRET与AIE协同效应。杂化物对大肠杆菌、金黄色葡萄球菌表现协同抗菌活性,自由基清除能力提升,溶血率<2%,血液相容性优异,在生物成像与生物医学方向具备应用潜力。

Visualizing hydroxyl radical by a dual-channel NIR fluorescence chemosensor within biosystem#

摘要:羟基自由基是生物体系中主要氧化物种,与多种疾病相关;但寿命极短、稳态浓度极低,实时检测难度大。本文设计合成双通道荧光探针HR-YT,用于生物体系内•OH灵敏监测。探针响应快、灵敏度高,检出限低至28 nM;细胞毒性低,适用于活细胞、植物组织、斑马鱼模型中外源性/内源性羟基自由基检测。还可用于β-拉帕醌刺激下,从肿瘤细胞中筛选非肿瘤巨噬细胞,在生物成像领域具备应用潜力。

Characterizing cellular receptor dynamics in live fluorescence imaging using stochastic modeling and machine learning#

摘要:细胞表面受体动力学调控信号传导与内吞,可通过全内反射荧光显微镜单颗粒追踪定量。本文开发可解释机器学习框架,结合轨迹几何特征与随机森林分类器,区分布朗运动、定向运动、分数布朗运动、奥恩斯坦-乌伦贝克运动、连续时间随机游走五类运动模式。应用于HIV共受体CCR5轨迹,激动剂刺激后受体动力学从间歇相关扩散向受限吸引驱动运动转变,与受体聚集和内化一致。该框架将随机运动模型与生物学细胞状态关联起来,可定量分析复杂受体动力学,广泛适用于膜蛋白活细胞成像研究。

High-Resolution Fluorescence Microscopy-Based 3D Encoding Using Polarity-Sensitive Dyes for Rapid Screening of Microplastics in Complex Environmental Samples#

摘要:微塑料是全球关注的新兴污染物,亟需快速监测手段。本文提出基于高分辨荧光显微镜的三维编码策略,采用三种极性敏感染料,实现复杂环境样本微塑料快速筛查。微塑料表面极性差异使吸附染料发生分子内电荷转移,产生不同荧光颜色;氢键与偶极相互作用带来差异化响应,辅助编码识别。采用自制便携荧光显微镜采集图像,RGB转换为CIE L*a*b* 构建3D编码库,显著提升微塑料与基质区分度。该方法可快速识别12种常见微塑料,与10种天然非塑料基质清晰分开;坐标重复性RSD=0.69-4.99%。加标样品与真实环境样品验证,结果与FTIR高度吻合。

Texture Analysis Using Indocyanine Green Fluorescence Measurement During Porcine Hypothermic Oxygenated Machine Perfusion#

摘要:开发低温机器灌注条件下器官活力评估新方法,对拓展心死亡供体器官池意义重大。提出基于ICG荧光图像纹理分析的定量方法,采用灰度共生矩阵(GLCM)的“对比度”特征定义评估指标h,即ICG流入200 s后的平均对比度。采用0 min与45 min热缺血猪肝,经低温氧合机器灌注保存,再自体血复灌评估。定性观察缺血肝灌注不均一,存在局部无荧光区。定量指标h与再灌注120 min门静脉阻力强正相关(r=0.70,p=0.015),与肝动脉阻力r=0.40存在相关性。该方法移植前即可预测缺血再灌注损伤带来的血管阻力。

Temporal variability of tryptophan-like fluorescence as a surrogate for microbial contamination and its association with flow components of karst spring discharge#

摘要:研究爱尔兰西部岩溶泉微生物污染时间变异与岩溶地下水系统不同流组分的关联。全年高频采集色氨酸样荧光(TLF)浓度时间序列作为微生物污染替代指标。流量历时曲线、主衰退曲线、傅里叶变换、连续与离散小波分析结果表明,该泉为三重孔隙岩溶系统,包含快速管道、中等裂隙、慢速基质流场。TLF变异与慢速/基流弱相关,与中速与快速流组分相关性更强;与大肠杆菌丰度强相关(Pearson r = 0.703),支持其作为微生物污染替代指标。TLF时间模式与含水层内部流动结构相关,降雨强度或补给量不能完全解释变异。

Integrated SARA–fluorescence analysis of crude oil with fraction-specific calibration#

摘要:原油组成复杂多变,需要快速可靠的分析方法用于炼厂优化与环境取证。荧光光谱灵敏、无损,但定量、族组分分析潜力未充分开发。本研究将SARA(饱和烃、芳烃、胶质、沥青质)族分离与系统荧光表征结合,表征伊拉克巴士拉原油。各族组分具有独特可重现的荧光特征:饱和烃与芳烃发射强度随浓度线性升高,R²>0.998;胶质与沥青质呈反比线性响应,R²>0.997,源于聚集诱导淬灭。建立族组分线性校准方程。350 nm激发下,发射峰从饱和烃(388 nm)、芳烃(435 nm)到沥青质(455 nm)、胶质(527 nm)系统红移,可作为多环芳烃缩合程度的光谱标尺。方法准确度90-105%,精密度日内RSD<5%;与GC-MS交叉验证无统计学偏差。

Quantifying the aging of dust aerosols using mie–polarization–fluorescence lidar: Linking optical evolution to chemical transformations#

摘要:沙尘颗粒在长距离传输中经历复杂物理化学老化,改变光学性质与环境影响。本文利用米散射-偏振-荧光激光雷达结合地面观测,研究中国西北沙尘混合与老化过程。构建沙尘老化指数(DAI),整合多光学参数量化混合过程与老化状态。荧光光谱的阶段变化反映老化过程中系统组成变化;新鲜沙尘混合以短波发射为主,混合老化后光谱峰向长波移动,对应高氧化有机组分贡献增加。DAI呈非单调演化,表明沙尘老化经历多个过渡阶段。进一步分析发现长波荧光发射、亚微米颗粒、二次气溶胶前体之间强相关,沙尘老化促进氧化有机物形成与积累。

Deep learning-based excitation-emission matrix fluorescence spectroscopic analysis of polycyclic aromatic hydrocarbons in coal tar#

摘要:煤焦油中多环芳烃(PAHs)快速准确检测至关重要,但光谱重叠与基质效应带来挑战。激发-发射矩阵(EEM)荧光光谱结合深度学习算法可应对该挑战。提取环己烷稀释煤焦油样品,构建覆盖3-6环PAHs与不同浓度梯度的117套EEM光谱数据集。七种机器学习算法用于定性分类,ResNet18深度学习算法准确率最高,接近90%。采用卷积神经网络变分自编码器模型定量分析,均方根误差RMSE低至0.0036 mg/L,平均绝对误差MAE低至0.0028 mg/L。该方法成本低、快速、可靠,可缓解瑞利散射与光谱重叠问题。

A fluorescence-based protocol for quantifying microplastics in soil: Protocol optimization and field investigation#

摘要:陆地环境微塑料累积加剧,亟需可靠的土壤定量方法。本研究建立并优化土壤微塑料定量方案,包括土壤有机质消解、微塑料提取、尼罗红染色、荧光显微镜检测、图像处理计数,重点优化消解与提取效率。评估H₂O₂、芬顿试剂、HNO₃三种消解试剂去除背景荧光有机质的能力,HNO₃背景荧光去除效率最高,壤土89%、黏土82%,且对聚丙烯、聚氯乙烯微塑料损伤小。CaCl₂与ZnCl₂提取效率相近,但CaCl₂环境毒性更低,PP回收率78-80%、PVC 61-70%。应用于三种不同土地利用类型土壤,农田微塑料浓度最高(1.04 × 10⁵个/kg),其次是填埋场与公园,<100 μm粒径占主导。

摘要:天然牙齿因牙本质内有机成分而天然发荧光,决定其活力与紫外下外观。当代树脂复合材料越来越多地掺入荧光剂模拟该特性,提升与天然牙列的美学融合。本综述考察天然牙与直接树脂复合材料的荧光特征,评估牙科荧光评估方法,识别影响荧光稳定性与仿生性能的因素。天然牙发射峰一般在410-500 nm,牙本质强度高于釉质。复合树脂通常掺入稀土氧化物荧光剂,发射波长相近(约450-485 nm),但强度因品牌与色度差异大。材料荧光受组成、老化与环境暴露影响。尽管荧光仿生取得进展,但材料间变异性仍然可区分天然牙与修复体,因此荧光方法仍可用于修复体识别。
